Carvacrol and Thymoquinone: Why Oregano Oil and Black Seed Oil Are Paired

Carvacrol and oregano oil

Oregano essential oil is what comes out of a still when oregano leaves and flowering tops are steam-distilled. Its composition is dominated by carvacrol, with smaller amounts of thymol, p-cymene and γ-terpinene, in proportions that depend on the plant. Because that variation is large, a serious supplier standardises the oil — blends or selects lots so that carvacrol lands at a stated level, commonly 60–80%. That standardisation figure is what "70% carvacrol" on a label refers to: the oregano oil ingredient, before dilution.

Undiluted, the oil is a strong irritant to skin and mucous membranes, which is why it is never sold neat for internal use. Every retail oil of oregano is a small quantity of the essential oil dispersed in a much larger quantity of a carrier oil. Most of what is written about oregano oil's properties is laboratory work on carvacrol: in vitro studies of how the compound behaves against microorganisms in culture, and animal studies of other properties. That literature is real and large; it is also a literature about an isolated compound at experimental concentrations, not about a dropper of diluted oil in a person.

Thymoquinone and black seed oil

Nigella sativa is an annual in the buttercup family whose seeds have been pressed for oil across the Middle East, North Africa and South Asia for a very long time. The oil's characteristic compound is thymoquinone, a quinone that is present at low single-digit percentages at best and that is sensitive to heat, light and time. That is why cold pressing matters for this oil in a way it does not for, say, sunflower oil, and why the thymoquinone content is something to document per batch rather than promise on a label. Why the two end up in one bottle

The pairing is a consequence of a formulation constraint, not a discovery. Oregano oil must be diluted into a carrier. The carrier is normally olive or sunflower oil, chosen for being cheap and neutral. If instead the carrier is cold-pressed black seed oil, then the roughly nine-tenths of the bottle that was going to be filler is now an oil with its own characteristic compound. That is the entire logic of an oil of oregano with black seed oil: the carrier is not wasted.

Two things follow from that. First, black seed oil is doing the carrier's job, so its own composition matters — an unrefined, cold-pressed, documented oil makes the pairing meaningful; a refined or old one does not. Second, the pairing is not evidence that the two compounds do anything together. Two rules cover most of it. A percentage attached to a compound describes the input oil unless the label says otherwise. And a figure in milligrams is only as good as the density and the batch document behind it; essential oils and seed oils are both slightly less dense than water, so a milligram figure calculated as if 1 mL weighed 1,000 mg overstates the content by several percent. Neither error is dishonest in itself, but a label that does not say which it is doing cannot be checked.

What the research on the two compounds does and does not show

Both compounds have been studied for decades, almost entirely as isolated substances in laboratory and animal models. That work is why they are interesting and why they are on labels.
